Dasatinib plus quercetin senolytic therapy reduces atrial fibrillation susceptibility in mice after cardiac irradiation
In mice given 20 Gy cardiac irradiation, treatment with dasatinib and quercetin reduced DNA damage, excessive autophagy, and GATA4-NF-κB signaling. The senolytic combination lowered senescence-associated secretory phenotype markers and reversed atrial structural and electrophysiological remodeling. Treated mice showed markedly reduced susceptibility to atrial fibrillation, a common complication after thoracic radiotherapy. The findings point to cellular senescence as a driver of radiation-induced atrial fibrillation and to senolytics as a possible prevention strategy.
